Psilocybin may prevent chemotherapy‑induced peripheral neuropathy
Chemotherapy‑induced peripheral neuropathy—characterised by numbness, tingling and pain in the hands and feet—affects a significant proportion of cancer patients and can lead to dose reductions or treatment discontinuation. Recent preclinical and early‑phase clinical studies have indicated that psilocybin, the psychoactive compound found in certain species of mushrooms, may mitigate these neurotoxic effects. In laboratory models, psilocybin administration reduced markers of nerve damage and improved sensory function in mice treated with neurotoxic chemotherapeutic agents.
The research, conducted by a multidisciplinary team of neuroscientists and oncologists, involved a randomized, double‑blind trial in which patients receiving standard chemotherapy received either a low‑dose psilocybin infusion or placebo. Preliminary results showed a statistically significant lower incidence of neuropathic symptoms in the psilocybin group, with patients reporting less pain and improved hand‑foot coordination. While the exact mechanism remains under investigation, hypotheses centre on psilocybin’s interaction with serotonin 5‑HT2A receptors and its potential to promote neuroplasticity and anti‑inflammatory pathways.
If these findings are confirmed in larger, multi‑centre trials, psilocybin could become a novel adjunct therapy to preserve quality of life and treatment adherence for cancer patients. Regulatory and safety considerations will need to be addressed, but the prospect of repurposing a naturally occurring psychedelic for a common chemotherapy side effect represents a promising avenue for improving supportive cancer care.
