Pirtobrutinib Adds Benefit to Venetoclax-Rituximab in CLL

A study presented at the European Hematology Association’s annual congress in Stockholm (June 11‑14) reported that adding the BTK inhibitor pirtobrutinib to venetoclax‑rituximab (PVR) improves progression‑free survival (PFS) in patients with relapsed or refractory chronic lymphocytic leukemia (R/R CLL) compared with venetoclax‑rituximab (VR) alone.

The analysis focused on adults with R/R CLL who had previously received therapy. Patients receiving the PVR combination experienced a longer period without disease progression than those treated with the standard VR regimen. The finding underscores the potential benefit of incorporating pirtobrutinib into existing chemo‑immunotherapy protocols for this difficult‑to‑treat population and suggests a new therapeutic option that may extend disease control.

If confirmed in larger, randomized trials, the PVR regimen could influence future treatment guidelines for R/R CLL, offering clinicians an additional strategy to prolong PFS and improve patient outcomes.
