New Clinical Trial Data Shows Promising Results for Rare Bile Duct Cancer Treatment
The eNRGy trial, a Phase II global study of zenocutuzumab in patients with solid tumors harboring NRG1 gene fusions, has demonstrated favorable safety and efficacy outcomes, particularly in those with advanced cholangiocarcinoma. The single-arm, multicenter trial reported that previously treated patients with NRG1-positive cholangiocarcinoma experienced a near doubling of progression-free survival compared to standard care expectations, alongside a manageable safety profile. These findings, published in a clinical oncology journal, highlight the potential of zenocutuzumab—a HER2/HER3 dual-targeting antibody—to address a significant unmet need in this rare and aggressive cancer subtype.
The trial enrolled patients with various NRG1-fusion-positive solid tumors, but the most notable results emerged in cholangiocarcinoma, a bile duct cancer with limited treatment options. Among 31 evaluable patients, median progression-free survival reached 7.2 months, exceeding historical benchmarks for second-line therapies in the disease. Adverse events were predominantly low-grade, with no unexpected toxicities observed. Researchers emphasized that these data support further investigation of zenocutuzumab, particularly in combination with other agents, to optimize outcomes for patients with NRG1-driven malignancies.
The results underscore the importance of molecular profiling in cancer care, enabling targeted therapies for genetically defined subsets. While the single-arm design limits direct comparison to standard treatments, the trial’s outcomes provide a rationale for advancing zenocutuzumab into Phase III studies. If confirmed in larger trials, the therapy could become a novel treatment option for NRG1-positive cancers, offering hope for improved survival and quality of life in a patient population with historically poor prognoses.
